  • 简介:Objective:ThisstudyaimstoexploretheassociationbetweenthedensityofHimalayanmarmot(Marmotahimalayana)andclimaticfactorssuchastemperature,precipitation,humidity,vapourpressure,sunshinepercentage,windvelocity,whicharecloselyassociatedwithglobalclimatechange,andtoprovideareferenceforplaguepreventionandcontrol.Methods:WeconductedaregressionanalysistofindthepossibleclimatefactorsassociatedwiththedensityofHimalayamarmot,andanalyzedtheresponsecharactersofHimalayanmarmottoclimatechange.Results:Dailyprecipitationdays(>=0.1mm)andsunshinepercentageweresignificantlyassociatedwiththedensityofHimalayanmarmot(p<0.01).Conclusion:Climatechangewasassociatedwiththeriskofplague.ThisphenomenonisvaluableforHimalayanmarmotandplagueprevention.MorestudiesareneededtounderstandtheimpactofclimatechangeonHimalayanmarmotandplague.

  • 简介:Carbonnanotubescancarryproteinintocellstoinducebiologicaleffects.Amino-functionalizedcarbonnanotubesaresolubleandbiocompatible,havehighreactivityandlowtoxicity,andcanhelppromotenervecellgrowth.Inthisstudy,amino-functionalizedethylenediamine-treatedmulti-walledcarbonnanotubeswereusedtopreparecarbonnanotubes-nervegrowthfactorcomplexesbynon-covalentgrafting.Thephysicochemicalproperties,cytotoxicitytoPC12andchickembryodorsalrootganglion,andbiologicalactivityofthecarbonnanotubes-nervegrowthfactorcomplexeswereinvestigated.Theresultsshowedthataminofunctionalizationimprovedcarbonnanotubes-nervegrowthfactorcomplexdispersibility,reducedtheirtoxicitytoPC12cells,andpromotedPC12celldifferentiationandchickembryodorsalrootganglion.

  • 简介:Hypoxia-induciblefactor1(HIF-1)attenuatesamyloid-betaproteinneurotoxicityanddecreasesapoptosisinducedbyoxidativestressorhypoxiaincorticalneurons.Inthisstudy,weconstructedarecombinantadeno-associatedvirus(rAAV)vectorexpressingthehumanHIF-1αgene(rAAV-HIF-1α),andtestedtheassumptionthatrAAV-HIF-1αrepresseshippocampalneuronalapoptosisinducedbyamyloid-betaprotein.OurresultsconfirmedthatrAAV-HIF-1αsignificantlyreducesapoptosisinducedbyamyloid-betaproteininprimaryculturedhippocampalneurons.DirectintracerebralrAAV-HIF-1αadministrationalsoinducedrobustandprolongedHIF-1αproductioninrathippocampus.SinglerAAV-HIF-1αadministrationresultedindecreasedapoptosisofhippocampalneuronsinanAlzheimer’sdiseaseratmodelestablishedbyintracerebroventricularinjectionofaggregatedamyloid-betaprotein(25–35).OurinvitroandinvivofindingsdemonstratethatHIF-1haspotentialforattenuatinghippocampalneuronalapoptosisinducedbyamyloid-betaprotein,andprovidesexperimentalsupportfortreatmentofneurodegenerativediseasesusinggenetherapy.

  • 简介:Homeoboxtranscriptionfactorsparticipateinthegrowthanddevelopmentofplantsbyregulatingcelldifferentiation,morphogenesisandenvironmentalsignalresponse.Torevealthefunctionsofthesetranscriptionfactorsinrice,weconstructedtheRNAivectorsofOsHox9,amemberofhomeoboxfamily,andanalyzedthefunctionofOsHox9usingreversegenetics.TheplantheightandtilleringnumberofRNAitransgenicplantsdecreasedcomparedwiththoseofwild-typeplants.Reversetranscription-polymerasechainreactionanalysisshowedthatOsHox9expressionreducedinthetransgenicplantswithphenotypicvariance,whereasthatinthetransgenicplantswithoutphenotypicvariancewassimilartothatinthewild-typeplants.ThisresultsuggeststhatthephenotypesofthetransgenicplantswerecausedbyRNAieffects.Thetissue-specificityofOsHox9expressionindicatedthatitwasexpressedindifferentorgans,withhighexpressioninstemapicalmeristemandyoungpanicles.SubcellularlocationofOsHox9demonstratedthatitwaslocalizedonthecellmembrane.

  • 简介:AIM:Toinvestigatethemorphologicalalteringeffectoftransforminggrowthfactor-β2(TGF-β2)onuntransfectedhumancornealendothelialcells(HCECs)invitro.METHODS:AfteruntransfectedHCECsweretreatedwithTGF-β2atdifferentconcentrations,themorphology,cytoskeletondistribution,andtypeIVcollagenexpressionofthecellswereexaminedwithinvertedcontrastlightmicroscopy,fluorescencemicroscopy,immunofluorescenceorWesternBlot.RESULTS:TGF-β2attheconcentrationof3-15μg/LhadobviouslyalterativeeffectsonHCECsmorphologyindoseandtime-dependentmanner,and9μg/Lwasthepeakconcentration.TGF-β2(9μg/L)alteredHCEcellmorphologyaftertreatmentfor36h,increasedthemeanopticaldensity(P<0.01)andthelengthofF-actin,reducedthemeanopticaldensity(P<0.01)ofthecollagentypeIVinextracellularmatrix(ECM)andinducedtherearrangementofF-actin,microtubuleincytoplasmandcollagentypeIVinECMaftertreatmentfor72h.·CONCLUTION:TGF-β2hasobviouslyalterativeeffectonthemorphologyofHCECsfrompolygonalphenotypetoenlargedspindle-shapedphenotype,indoseandtime-dependencemannerbyinducingmore,elongationandalignmentofF-actin,rearrangementofmicrotubuleandlargerspreadareaofcollagentypeIV.

  • 简介:Neuroprotectionbyischemicpreconditioninghasbeenconfirmedbymanystudies,buttheprecisemechanismremainsunclear.Inthepresentstudy,weperformedcerebralischemicpreconditioninginratsbysimulatingatransientischemicattacktwice(eacha20-minuteocclusionofthemiddlecerebralartery)beforeinducingfocalcerebralinfarction(2hourocclusion-reperfusioninthesameartery).Wealsoexploredthemechanismunderlyingtheneuroprotectiveeffectofischemicpreconditioning.Sevendaysafterocclusion-reperfusion,tetrazoliumchloridestainingandimmunohistochemistryrevealedthattheinfarctvolumewassignificantlysmallerinthegroupthatunderwentpreconditioningthaninthemodelgroup.Furthermore,vascularendothelialgrowthfactorimmunoreactivitywasconsiderablygreaterinthehippocampalCA3regionofpreconditionedratsthanmodelrats.Ourresultssuggestthattheprotectiveeffectsofischemicpreconditioningonfocalcerebralinfarctionareassociatedwithupregulationofvascularendothelialgrowthfactor.

  • 简介:Endothelialprogenitorcellsareresidentinthebonemarrowbloodsinusoidsandcirculateintheperipheralcirculation.Theymobilizefromthebonemarrowaftervascularinjuryandhometothesiteofinjurywheretheydifferentiateintoendothelialcells.Activationandmobilizationofendothelialprogenitorcellsfromthebonemarrowisinducedviatheproductionandreleaseofendothelialprogenitorcell-activatingfactorsandincludesspecificgrowthfactorsandcytokinesinresponsetoperipheraltissuehypoxiasuchasafteracuteischemicstrokeortrauma.Endothelialprogenitorcellsmigrateandhometospecificsitesfollowingischemicstrokeviagrowthfactor/cytokinegradients.Somegrowthfactorsarelessstableunderacidicconditionsoftissueischemia,andsyntheticanaloguesthatarestableatlowpHmayprovideamoreeffectivetherapeuticapproachforinducingendothelialprogenitorcellmobilizationandpromotingcerebralneovascularizationfollowingischemicstroke.

  • 简介:TheresistivityexperimentalmeasurementsofcoresamplesdrilledfromlowpermeabilityreservoirsofOrdosBasin,NorthwestChina,illustratethatthecementationfactorsarenotagminate,butvaryfrom1.335to1.749.Thisleadstoachallengefortheestimationofwaterandhydrocarbonsaturation.BasedontheanalysisofPurcellequationandassumptionthatrockresistivityisdeterminedbytheparallelconnectionofnumerouscapillaryresistances,atheoreticalexpressionofcementationfactorintermsofporosityandpermeabilityisestablished.Then,cementationfactorcanbecalculatediftheparametersofporosityandpermeabilityaredetermined.Inthefieldapplication,porositycanbeeasilyobtainedbyconventionallogs.However,itisatoughchallengetoestimatepermeabilityduetothestrongheterogeneityoflowpermeabilityreservoirs.Thus,theSchlumbergerDollResearch(SDR)modelderivedfromNMRlogshasbeenproposedtoestimatepermeability.BasedontheanalysisofthetheoreticalexpressionsofcementationfactorandSDRmodel,anovelcementationfactorpredictionmodel,whichisrelevanttoporosityandlogarithmicmeanofNMRT2spectrum(T2lm),isderived.TheadvantageofthismodelisthatalltheinputinformationcanbeacquiredfromNMRlogsaccurately.Inordertoconfirmthecredibilityofthenovelmodel,theresistivityandcorrespondinglaboratoryNMRmeasurementsof27coresamplesareconducted.Thecredibilityofthemodelisconfirmedbycomparingthepredictedcementationfactorswiththecoreanalyzedresults.Theabsoluteerrorsforallcoresamplesarelowerthan0.071.Oncethismodelisextendedtofieldapplication,theaccuracyofwaterandhydrocarbonsaturationestimationwillbesignificantlyimproved.
