Objective:TodirectionallyclonetheomplgenefromChlamydiatrachomatis(Ct)FGenotypeontoaplasmidvectorforconstructingarudimentaryDNAvaccine.Methods:ThecompleteomplgenefromgenomicDNAofCtFgenotypewildspecieswasamplifiedwithprimersdesignedbycomputer.Therecombinantgenewasobtainedbyrestrictionenzymecutting,linkingthegenewiththeplasmidvectorinvitro,transformingtherecombinantgeneintobacteria,andextractingtheDNAfromthebacteria.Results:DNAextractingfromthebacteriawascomposedoftheimplgeneandplasmid,whichisidentifiedbythreemethodsofsingularrestrictiveenzymecutting,doublerestrictiveenzymecuttingandPCR.Conclusion:CloningoftheomplgenefromtheCtFgenotypemeansthatarudimentaryDNAvaccinewassuccessfullyconstructed.
