Modulation of signal transduction pathways by natural compounds in cancer

摘要 Cancerisgenerallyregardedastheresultofabnormalgrowthofcells.AccordingtoWorldHealthOrganization,canceristheleadingcauseofmortalityworldwide.Mothernatureprovidesalargesourceofbioactivecompoundswithexcellenttherapeuticefficacy.Numerousphytochemicalsfromnaturehavebeeninvestigatedforanticancerproperties.Inthisreviewarticle,wediscussseveralnaturalcompounds,whichhaveshownanti-canceractivity.Naturalcompoundsinducecellcyclearrest,activateintrinsicandextrinsicapoptosispathways,generateReactiveOxygenSpecies(ROS),anddown-regulateactivatedsignalingpathways,resultingininhibitionofcellproliferation,progressionandmetastasisofcancer.Severalpreclinicalstudieshavesuggestedthatnaturalcompoundscanalsoincreasethesensitivityofresistantcancerstoavailablechemotherapyagents.Furthermore,combiningFDAapprovedanti-cancerdrugswithnaturalcompoundsresultsinimprovedefficacy.Onthebasisoftheseexcitingoutcomesofnaturalcompoundsagainstseveralcancertypes,severalagentshavealreadyadvancedtoclinicaltrials.Inconclusion,preclinicalresultsandclinicaloutcomesagainstcancersuggestpromisinganticancerefficacyofagentsfromnaturalsources.
